In order to identify a misfiring cylinder, I know it is common practice to disconnect each HT lead one by one.

Is it best to do this at the distributor or directly off the top of the spark plug?

How dangerous is it to do with the engine running (I know this is the ideal case but I'm almost certainly going to electrocute myself)?

TIA as always :)

take it off the spark plug, I would advise turning the engine off whilst doing so !

Safe as long as you use a rag or rubber/dry cloth gloves to insulate yourself.

It's not necessarily dangerous (unless you have a heart condition or pacemaker fitted) to do it with bare hands, but it may well give you a nasty jolt, so best to ensure that you're insulated.

Pull a spark plug lead off of a running engine, or start one with a lead off, and you run the risk of 'spiking' the ECU when the EHT voltage goes through the roof and flashes over.
